Freshmans late goal lifts Ohio State womens soccer team into Sweet 16

Credit: Courtesy of OSUThe Ohio State women’s soccer team (13-6-3) is headed to the Sweet 16 after defeating the Virginia Tech Hokies (15-4-2) on Friday afternoon 1-0 in the second round of the NCAA tournament. Freshman midfielder Arden Holden netted her first career goal with 11 minutes remaining to keep the Buckeyes’ postseason run afloat. With the win, the Scarlet and Gray are headed to their third Sweet 16 in the past six seasons. The beginning of the game was slow, as the first 15 minutes did not present many opportunities to score for either side. In the 20th minute, OSU finally got to a solid opportunity in the final third when senior midfielder/forward Michela Paradiso tried to curl one in from 20 yards out on the left wing but it sailed over the crossbar. Two minutes later, the Buckeyes had another opportunity to score when junior forward Lindsay Agnew sent a strong ball to junior forward Nichelle Prince, but a Virginia Tech defender cleared it away. The Hokies, who entered the game as the 14th-overall seed, were able to generate a scoring opportunity in the 43rd minute when sophomore forward Alani Johnson sent one on the frame, but OSU redshirt junior goalkeeper Jillian McVicker made a lunging save to preserve the scoreless match. The teams headed into the locker rooms at halftime tied 0-0. The Hokies led the Buckeyes 6-1 in shots, 1-0 on goal. OSU came back in the second half with plenty of opportunities to score early on, but the Buckeyes simply could not find the back of the net. In the 79th minute, the Buckeyes put the one and only goal on the board. Prince split two Virginia Tech defenders before playing the ball out to Holden who finished off the keeper. Holden has appeared in 21 of the Buckeyes’ 22 games this season, but her game-winning goal marked the first time in her young career she has scored. The final 11 minutes of play contained some opportunities for the Hokies to level the match, including a solid look off a free kick in the 87th minute, but they could not get past McVicker, meaning OSU was able to hold on for the 1-0 win. For the game, Virginia Tech held a sizable lead in shots, 20-5, including 4-2 on goal. McVicker made four saves to earn her second clean sheet of the season for OSU. The Buckeyes are set to play conference foe Penn State in the Sweet 16 after the Nittany Lions dominated Boston University in the second round, winning 6-0. In the lone meeting between the two teams during the regular season, Penn State topped OSU 2-1 on Oct. 24. David Jaffe, with the Broad Institute reported to an audience at the Advances in Genome Biology and Technology meeting in Florida last week that he has been asked by Oxford Nanopore to try out the new the device, called the MinION, and has found it to be promising. Explore further Oxford Nanopore Technologies created a stir in the biological sciences world in 2012 when representatives for the company announced that its research team had successfully created a sequencing device based on nanopore technology and that an inexpensive prototype would be delivered to researchers outside the company soon thereafter. The company apparently ran into difficulty ramping up its pore technology and had to find some alternative materials—thus the two year delay.Nanopore sequencing is where single strands of DNA are pulled through a pore—as each base pair passes through the pore its conductivity is measured—information that can be used for identification by a computer. The advantage of this approach is that at least in principle, any strand length can be sequenced.The MinION isn’t for sale yet—at this time the company is making prototypes available to a select few notables in the genome sequencing field—each will have to pony up $1000 as a down-payment for the honor. The device plugs into a port on a computer, which does the processing. Along with the device, which has been compared to the size of a pack of gum, Oxford Nanopore Technologies will send along DNA samples that researchers can use to learn how to use the new device—after that, they can sequence anything they want. In testing strands of two types of bacterial DNA sent to him, Jaffe reports having mixed results —long stretches of data were obtained correctly in some cases, but in others there were some errors. In order to assemble the entire genome of a type of bacteria, for example, he had to also use a sequencing device made by another company. He noted that he is optimistic about the device, however, as reps from Oxford Nanopore have assured him that error rates can be reduced using certain techniques even as improvements to the device are being worked out back at the lab.If the error rates using the MinION can be reduced to a practical level, the device could mark a turning point in gemone sequencing as it would be a device that could be carried and used for fieldwork. And though the cost of such a device has not been announced, Oxford Nanopore has repeatedly used terms such as inexpensive and affordable to describe it, indicating it will cost much less than other sequencing devices currently on the marker, once it’s ready for sale. They found that the NHC bound to gold forms an adatom that then traverses the gold surface in a ballbot-type motion. Their report appeared in a recent issue of Nature Chemistry.NHCs form a particularly strong bond with gold because of the sp2 hybridized lone pair of electrons on the carbon atom (See figure of NHCs above). Studies have shown that NHCs can be used to form particularly stable self-assembled monolayers (SAMs) on gold surfaces and can even be modified after formation of the monolayer, but these studies are difficult to compare with each other because they do not have similar R groups.The nitrogen-substituted R groups play a significant role in the behavior of the NHCs. Bulky R groups prohibit dimerization but may affect the stability of monolayer formation. Also because the nitrogen atoms are part of an aromatic ring, they serve as electron donors to the substituents. The type of substituent affects the electronics of the molecule.In this study, Wang et al. used three different NHCs that were synthesized in the Glorius group, each of which had R groups that had previously been studied and represented a range of steric and electronic properties. These groups include IMes which has 2,4,6 trimethyl phenyl rings as the N-substituents; IPr which has 2,6-diisopropylphenyl substituents; and IMe which just has methyl groups as the N-substituents. This indicated that the surface was mobile, but there was no change to morphology of the gold surface. Despite this mobility, the bond between the NHC and the gold surface proved to be highly stable at high temperatures.This behavior may be explained by the formation of adatoms, or the interaction of one NHC molecule with one gold atom. This adatom then moves around the surface as one entity, which the authors liken to ballbot movement. They confirmed this structure by looking at the height of the IPr monolayers.DFT calculations helped to explain how exactly these adatoms formed and moved. IPr and IMe likely bound to a gold atom that was pulled from the surface. Simultaneously, a gold atom from the second layer moves into the vacant hole. The gold atom that was pulled from the surface was bound irreversibly to the NHC to form an adatom that then readily moved across the surface. Additional free energy calculations looked at the effects of bulky R groups, specifically comparing IPr with IMes. Wang et al. found that the pi interactions between the phenyl rings and the surface play a role in adatom formation and mobility. They reasoned that there is a balance between steric hindrance, as with IPr, and pi interactions, as displayed with IMes. And, indeed, the non-bulky IMe compound, which was more mobile than IPr and IMes, was able to form dimeric and trimeric complexes with one gold atom.This study provides important insights into how NHC-Au monolayers form and behave. They seem to form a stronger bond than thiols with gold, but they also display mobility that is influence by the identity of the nitrogen substituents. The NHCs that tend to have weaker pi interacts with the gold surface readily forming adatoms that then move across the surface like a ballbot. These NHCs extract a gold atom from the surface, which leads to the formation of an NHC–gold adatom complex that displays a high surface mobility by a ballbot-type motion.
